He started in 1962, so it's certainly possible that Fleming would have said this. IIRC, Fleming always said that he had Hoagy Carmichael in mind when describing Bond, so there's that.
It's hard to pick one Bond film as the best.
Casino Royale and
Skyfall would both be toward the top of the list for their depth and realism; the first three Connery films (
Dr. No, From Russia with Love, and
Goldfinger) would be up there too for establishing the icon. I'd be tempted to include
On Her Majesty's Secret Service up there, too, in spite of the Lazenby hate. Diana Rigg is probably my favorite Bond girl (though Eva Green is right up there), but Telly Savalas kinda sucks. Jack Lord from
Dr. No is my favorite Felix Leiter hands down.
Best Villain? Gotta be Scaramanga. Let's face it: Christopher Lee effectively controls the universe by now.
