Anyone see Pandemonium? It's from the 80s. It's along the same lines of the Scary Moviespoofs of today. The movie had quite a few big stars in it, although I'm not sure if they were big stars upon release (Phil Hartman, Paul Reubens, Judge Reinhold.) Pretty sure Tom Smothers was pretty famous already, another cast member.

Rat Race omg, I loved the guy with the tongue piercing best! Hilarious movie!!
(Actually, I enjoy most anything with Seth Green.)

Sugar and Spice

Mrs. Doubtfire

My Big Fat Greek Wedding I usually don't like romantic comedies, but this one charmed me to the core!

The Chevy Chase Vacation movies

White Chicks

The House Bunny

All in all, the sillier=the better for me!

Just because the style of humor is different than most of the movies posted here I have to add In Bruges. I thought it was hilarious but would not recommend watching the trailer.

Rather than repeating some of those already mentioned that I thoroughly enjoyed, I'll add a few that -- somewhat surprisingly -- have not been mentioned.

Love and Death -- Woody Allen's version of the great Russian novels.

Pocketful of Miracles -- combination romantic comedy/gangster movie by Frank Capra. Ann Margaret isn't that impressive, and overall not one of the best, but I give it extra credit for Bette Davis as well as great character actors like Thomas Mitchell, Peter Falk, Arthur O'Connell, and Edward Everett Horton.

The Cheap Detective -- Peter Falk in a Neil Simon parody of Bogey. Although I enjoy Neil Simon, I rarely include his movies among the best, but this is one of my favorites of all time. You may never been able to watch Casablanca, The Maltese Falcon, The Big Sleep, To Have and Have Not, etc. with a straight face again.

Default Funniest board room moment ever..

Jim Carrey in "Liar, Liar" and his 2 minutes of honest reflection in the board room is the loudest I have ever laughed in a theater... enjoy!

Ah, but we need to extend our horizons here. Both overseas ('La Cage aux Folles', far better than the American remake, and "Bedazzled," with Raquel Welch unforgettable as 'Lust') and back in time. The American Film Institute lists "Some Like It Hot" as the best US comedy of all time, and it certainly needs serious (!) consideration. But early Marx Brothers, ("Duck Soup", "A Night at the Opera,") Laurel & Hardy ("Way Out West"), Chaplin ("The Gold Rush",) Buster Keaton ("The General",) Mae West ("She Done Him Wrong",) and W.C. Fields ("Never Give a Sucker an Even Break",) are all top notch.
